Isothermal Titration Calorimetry for Optimizing Leads

Unlike traditional "affinity only" methods, label-free ultrasensitive microcalorimetry gives you a complete picture of a biomolecular interaction, including insights into the binding mechanism that are unavailable by other methods.

Using Isothermal Titration Calorimetry (ITC), you can quickly:

  • Determine a gold standard Kd values along with binding mechanism and stochiometry
  • Measure binding in a biologically relevant environment
  • Minimize assay development
  • Assess binding mechanism unconstrained of assay limitations

ITC applications areas include:

  • Hit validation
  • Fragment-based screening
  • Lead optimization
  • Mechanism of action studies

Now, the fully-automated Auto-iTC200 makes it possible to obtain complete binding profile with reduced sample consumption at throughput rates suitable for secondary screening.  The MicroCal Auto-iTC200 is designed to address the needs of today’s life science researchers — particularly those in drug discovery and development. The system combines the performance of MicroCal's ultra-sensitive iTC200 with full automation for a complete solution when unattended operation is desired.

Completely automated, all functions are operated through software. The software includes integrated design wizards to assist in selecting experimental run parameters.

A range of applications can be accomplished with the Auto-iTC200, including: characterization of molecular interactions of small molecules, proteins, antibodies, nucleic acids, lipids and other biomolecules, enzyme kinetics and the effects of molecular structure changes on binding mechanims.
